Routine Sickle Test

Description

When HbS is present in the reduced form it is insoluble in a phosphate buffer. The test involves adding a small volume of whole blood to a phosphate buffer containing sodium dithionite as a reducing agent and saponin as a lysing agent. As HbS is less soluble than normal adult haemoglobin in the presence of the reducing agent the solution remains cloudy whilst in normal patients the solution becomes clear.


Indications

A sickle solubility test will be performed in all instances where the primary Haemoglobinopathy screening method (Capillary Electrophoresis) detects a structural Hb variant.
